cDNA synthesis (complementary DNA synthesis) is the process of transforming RNA molecules existing in samples into DNA in complementary sequences. cDNA synthesis enables researchers to convert RNA into stable cDNA, which can be used for downstream applications such as PCR, microarray, and next-generation sequencing. By studying the amount of cDNA produced by genes, researchers can determine how much mRNA is present in the original sample and determine the expression level of specific genes.
In cDNA synthesis, reverse transcriptase is first used to reverse transcribe RNA samples into complementary DNA. The reverse transcriptase enzyme makes a complementary DNA copy of the RNA molecules present in the sample. cDNA synthesis is usually carried out using a mixture of random hexamer primer, which prime the reverse transcriptase to begin synthesizing from a various sites, and oligo-dT primers that anneal to the poly-A tail existing on most mRNAs.
